As the sentinels of innate and adaptive immune system, dendritic cells (DCs) have been considered to hold a great promise for medical application. Among diverse types DCs, monocyte-derived DCs (mo-DCs) generated in vitro most commonly employed. We improving culture protocol devised produce mature interferon-α-induced (IFN-DCs), hereinafter called (mat)IFN-DCs. While exploring relationship between expression CD56 cytotoxic activity (mat)IFN-DCs, we unexpectedly found that sorting (mat)IFN-DCs with antibody-coated microbeads (MB) resulted fractionating tumoricidal into flow-through (FT) but not MB-bound fraction. uncovered FT fraction contains expressing low substantial level CD56. Moreover, those express granzyme B (GrB), perforin (PFN), serpin B9 at high levels. By employing specific inhibitor PFN, confirmed direct relies on GrB/PFN pathway. designated subpopulation as CD56dim positively sorted CD56bright, respectively.
